What did Bruce Lee mean, does practice of one technique really make it deadly?

Well, Bruce Lee was a brilliant philosopher with many quotes that had deep and thoughtful feelings.

The above quote doesn't refer to a guy that has simply trained 1 kick 10,000 times but a guy that has dedicated not hours, days or weeks but years to training one simple tool; such as a kick or a punch.

If you practice something 10,000 times you will drill it into your muscle memory, work out all the details of alignment and geometry - then it will be a real weapon in your arsenal.

Therefore, a martial artist that has dedicated years of training a few effective tools, and trained them to work in almost any situation is considered far more dangerous than a martial artist that has dedicated years of training to hundreds of tools - not because all of them are useless but because there just isn't enough time in one lifetime to perfect each one.

Perfect practice of one thing makes oneself expert in that field. It doesn't matter how many books you study for a subject; if you read one book thoroughly, you will know the topic very well. It’s just like jack of all trades, master of none.
